The castle, understood as a fortified inhabited nucleus, established itself already in the 13th century with a significant demographic growth and in 1278 it was found that it had 150 fires inside.

After a period of dependence on the Abbey of Sant'Antimo during the 1200s Camigliano was attracted into the Sienese orbit and in the following years the representatives of some of the major city families (Gallerani, Bonsignori) implemented a strong neo-noble patrimonial expansion destined to supplant the traditional secular and ecclesiastical presences and to compress the nascent affirmation of small peasant property.

The noble presence is also felt in Camigliano which passed under the hegemony of these families for many years during the 13th century but the fact remains that the local community will never disappear completely from the populous Castle of Camigliano, where the strong component rural seems to have maintained its vitality for a few centuries.

The current manor house was built inside the access door (called "borgone") of the ancient "castle" taking advantage of the volume of the ancient walls that surrounded the town. The symbol of Camigliano: the camel, found in a 13th century seal, is perhaps due to the influence of the papacy in the area, and is hypothesized to be linked to the movements of the crusades that reached the holy land.
